RESUMO

In the traditional dry etching process for photonic device fabrication, the etching effect is influenced in many ways, usually resulting in relatively large sidewall roughness and high transmission loss. In this study, an effective method, namely the secondary coating method, is proposed to reduce the transmission loss of a Ge-Sb-Se chalcogenide waveguide and increase the quality factor (Q-factor) of a Ge-Sb-Se chalcogenide micro-ring resonator. The Ge-Sb-Se waveguide and micro-ring resonator are fabricated by ultraviolet exposure/electron beam lithography and inductively coupled plasma etching technology. Afterward, a 10 nm-thick Ge-Sb-Se thin film is deposited by thermal evaporation. The measurements show that after secondary coating, the sidewall roughness of the waveguide is reduced from 11.96 nm to 6.52 nm, with the transmission loss reduced from 2.63± 0.19 dB/cm to 1.86± 0.11 dB/cm at 1.55 µm wavelength. Keeping an equal coupling condition with equal radius and coupling distance, the Q-factor of the micro-ring resonator is improved by 47.5% after secondary coating. All results indicate that the secondary coating method is a feasible way to generate low-loss and high Q-factor integrated chalcogenide photonic devices.

RESUMO

Cap RNA methylations play important roles in the replication, evasion of host RNA sensor recognition, and pathogenesis. Coronaviruses possess both guanine N7- and 2'-O-ribose methyltransferases (N7-MTase and 2'-O-MTase) encoded by nonstructural protein (nsp) 14 and nsp16/10 complex, respectively. In this study, we reconstituted the two-step RNA methylations of N7-MTase and 2'-O-MTase of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in vitro and demonstrated its common and different features in comparison with that of SARS-CoV. We revealed that the nsp16/10 2'-O-MTase of SARS-CoV-2 has a broader substrate selectivity than the counterpart of SARS-CoV and can accommodate both unmethylated and uncapped RNA substrates in a sequence-independent manner. Most intriguingly, the substrate selectivity of nsp16/10 complex is not determined by the apoenzyme of nsp16 MTase but by its cofactor nsp10. These results provide insight into the unique features of SARS-CoV-2 MTases and may help develop strategies to precisely intervene in the methylation pathway and pathogenesis of SARS-CoV-2.

Rare-earth doped chalcogenide films are major components in flexible and integrated photonic and optoelectronic devices for modern communication systems, metrology, and optical sensing. However, it is still challenging to develop a high concentration of rare-earth doping chalcogenide film with a smooth surface to realize efficient photoluminescence (PL). Here, we demonstrate that Er3+-doped GeS2 films are prepared by spin-coating based on a two-step dissolution process. Such a two-step process provides the high solubility of Er3+ in GeS2 films and exhibits efficient emission at ∼1.5 µm crossing the telecommunication C-band. The highest PL emission intensity is obtained in GeS2 films doped with 1.4 mol% of Er3+, and this PL in GeS2 films is reported for the first time. We propose adjustments of annealing parameters for improving the PL characteristics in such materials. Through the control precision of the heating rate and annealing temperature, the smooth surface of GeS2 films enables efficient photo-luminescence. This two-step dissolution-based strategy would pave a new path to design luminescent chalcogenide films for application in flexible and integrated optoelectronics and photonics.

Surface-enhanced Raman scattering (SERS) has great potential in the field of rapid detection of pesticide residues in food. In this paper, a fiber optic SERS sensor excited by evanescent waves was proposed for efficient detection of thiram. Silver nanocubes (Ag NCs) were prepared as SERS active substrates, which had much stronger electromagnetic field intensity than nanospheres under laser excitation due to much more "hot spots". By using the method of electrostatic adsorption and laser induction, Ag NCs were uniformly assembled at the fiber taper waist (FTW) for enhancing the Raman signal. Different from the traditional way of stimulation, evanescent wave excitation greatly increased the interaction area between the excitation and analyte, while reducing the damage of the excited light to the metal nanostructures. The methods proposed in this work have been successfully used to detect the pesticide residues of thiram and showed good detection performance. The detection limits for 4-Mercaptobenzoic acid (4-MBA) and thiram were determined to be 10-9 and 10-8 M, the corresponding enhancement factor could be 1.64 × 105 and 6.38 × 104. Low concentration of thiram was detected in the peels of tomatoes and cucumbers, indicating its feasibility in actual sample detection. The combination of evanescent waves and SERS provides a new direction for the application of SERS sensors, which had great application potential in the field of pesticide residue detection.

S-217622 (Ensitrelvir) is a reversible severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) 3-chymotrypsin-like protease (3CLpro) inhibitor which obtained emergency regulatory approval in Japan for the treatment of SARS-CoV-2 infection on Nov 22, 2022. Herein, analogs of S-271622 with deuterium-for-hydrogen replacement were synthesized for comparison of the antiviral activities and pharmacokinetic (PK) profiles. Compared to the parent compound, C11-d2-S-217622 compound YY-278 retained in vitro activity against 3CLpro and SARS-CoV-2. X-ray crystal structural studies showed similar interactions of SARS-CoV-2 3CLpro with YY-278 and S-271622. The PK profiling revealed the relatively favorable bioavailability and plasma exposure of YY-278. In addition, YY-278, as well as S-217622, displayed broadly anti-coronaviral activities against 6 other coronaviruses that infect humans and animals. These results laid the foundation for further research on the therapeutic potential of YY-278 against COVID-19 and other coronaviral diseases.

In this article, a compact racetrack double microring resonator (MRR) sensor based on Ge28Sb12Se60 (GeSbSe) is investigated. The sensor device consists of a racetrack microring, an embedded small microring, and a strip waveguide. Electron beam lithography (EBL) and dry etching are used to fabricate the device. The compact racetrack double MRR device are obtained with Q-factor equal to 7.17 × 104 and FSR of 24 nm by measuring the transmission spectrum. By measuring different concentrations of glucose solutions, a sensitivity of 297 nm/RIU by linear fitting and an intrinsic limit of detection (iLOD) of 7.40 × 10-5 are obtained. It paves the way for the application of chalcogenide glasses in the field of biosensing.

A dual-peak long period fiber grating (DP-LPFG) sensor functionalized by polypyrrole-chitosan composite was proposed for sensitive detection of Cu2+ ions in aqueous solution. The nitrogen atom on the polypyrrole ring and the amino group on the chitosan chain in the complex matrix can chelate the Cu2+ ions. Thus, the refractive index of the overlay changed and further modulated the transmission spectrum of DP-LPFG. After special design, the double peaks can move in opposite directions with the increase of Cu2+ ion concentration, thereby greatly improving detection sensitivity. The linear sensitivity of the fabricated sensor was measured to be 9.12 and 2.14 nm/ppm (0.61 and 0.14 nm/µM) for concentrations of 0.1-0.5 (1.5 µM-7.5 µM) and 0.5-2 ppm (7.5 µM-30 µM), respectively. In addition, the Langmuir isothermal model was used to evaluate the overall response of the sensor to Cu2+ ions quantitatively, and the detection limit was determined to be 0.05 ppb (0.75 nM). This ingenious sensor offers a new solution for sensitive detection of heavy metal ions in environmental water.

Chalcogenide solid-state electrolytes (SEs) have been regarded as promising candidates for lithium dendrite suppression due to their high ionic conductivity, suitable mechanical strength, and large Li+ ion transference number. However, the wide applications of SEs in pragmatic all-solid-state batteries are still retarded by their limited interface stability, which leads to lithium dendrite growth and formation of interphase with high resistance. In addition, the interphase evolution mechanism between SEs and metallic Li anodes remains unclear. Herein, this work demonstrates that the interfacial stability of Li2S-SiS2-P2S5 SEs can be effectively enhanced by tuning the interphase through LiI incorporation. This strategy contributes to a high ionic conductivity of the SEs and electronic insulation interphase containing LiI. Thus, the 70(60Li2S-28SiS2-12P2S5)-30 LiI SEs prepared by melt-quenching exhibit a high ionic conductivity of 1.74 mS cm-1 at room temperature and a larger critical current density of 1.65 mA cm-2 at 65 °C. The cycling life of the symmetric Li|SEs|Li cell is up to 200 h without significant resistance growth at 0.1 mA cm-2 at room temperature. This enhanced interface stability is revealed to originate from the in situ-formed LiI within the interphase, which prevents continual SEs degradation and suppresses lithium dendrite growth. This work provides a vital understanding of interphase evolution, which is valuable for designing SEs with long cycling stability.

A sensitive ammonia sensor based on long-period fiber grating (LPFG) is designed and manufactured for the detection of ammonia concentration in water. Femtosecond laser direct writing technology is used to write LPFGs on standard single-mode silica fiber. A thin layer doped with basic dyes is coated on the optical fiber for sensing by using the sol-gel method. The thicknesses of sol-gel layers, which play a key role in the sensitivity of the LPFG sensor, were carefully studied. Experimental results show that LPFG with a functional layer of ∼340 nm has the best sensing performance, and the detection limit is 0.08 ppm. The response time of the sensor is less than one minute, and the sensor has good repeatability with a short recovery time. Compared with other organic molecules and ions in water, the proposed LPFG sensor has not only good reusability, but also selectivity for the detection of ammonia.

The nonlinear optical properties of emerging metal halide perovskites (MHP) materials are sufficiently intriguing that this topic has become the hotspots in the realm of material science. Hence, we investigate the third-order nonlinear optical properties of CsPbBrx/I3-x (x = 1, 2, 3) MHP nano-crystals (NCs) embedded chalcogenide glass (ChG) within a GeS2-Sb2S3 pseudo-binary system, by monitoring the composition, excitation wavelength and intensity dependencies via femtosecond Z-scan technique. We have found that the intrinsic large optical nonlinearity of ChG can be further enhanced because of the incorporation of MHP NCs, and that the optical nonlinearity of MHP-ChG containing pristine Br NCs is more pronounced compared to its counterparts with mixed Br/I NCs, due to a combination of multiple factors.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was initially described to target the respiratory system and now has been reported to infect a variety of cell types, including cardiomyocytes, neurons, hepatocytes, and gut enterocytes. However, it remains unclear whether the virus can directly infect human embryonic stem cells (hESCs) or early embryos. Herein, we sought to investigate this question in a cell-culture system of hESCs. Both the RNA and S protein of SARS-CoV-2 were detected in the infected hESCs and the formation of syncytium was observed. The increased level of subgenomic viral RNA and the presence of dsRNA indicate active replication of SARS-CoV-2 in hESCs. The increase of viral titers in the supernatants revealed virion release, further indicating the successful life cycle of SARS-CoV-2 in hESCs. Remarkably, immunofluorescence microscopy showed that only a small portion of hESCs were infected, which may reflect low expression of SARS-CoV-2 receptors. By setting |log2 (fold change)| > 0.5 as the threshold, a total of 1,566 genes were differentially expressed in SARS-CoV-2-infected hESCs, among which 17 interferon-stimulated genes (ISGs) were significantly upregulated. Altogether, our results provide novel evidence to support the ability of SARS-CoV-2 to infect and replicate in hESCs.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the virus driving the ongoing coronavirus disease 2019 (COVID-19) pandemic, continues to rapidly evolve. Because of the limited efficacy of vaccination in prevention of SARS-CoV-2 transmission and continuous emergence of variants of concern (VOCs), orally bioavailable and broadly efficacious antiviral drugs are urgently needed. Previously, we showed that the parent nucleoside of remdesivir, GS-441524, has potent anti-SARS-CoV-2 activity. Here, we report that esterification of the 5'-hydroxyl moieties of GS-441524 markedly improved antiviral potency. This 5'-hydroxyl-isobutyryl prodrug, ATV006, demonstrated excellent oral bioavailability in rats and cynomolgus monkeys and exhibited potent antiviral efficacy against different SARS-CoV-2 VOCs in vitro and in three mouse models. Oral administration of ATV006 reduced viral loads and alleviated lung damage when administered prophylactically and therapeutically to K18-hACE2 mice challenged with the Delta variant of SARS-CoV-2. These data indicate that ATV006 represents a promising oral antiviral drug candidate for SARS-CoV-2.

Bragg grating (BG) filters play important roles in integrated photonics such as signal processing and optical sensing. In silicon-based counterpart photonic platforms, the application of narrow-bandwidth (Δλ) filters is often restrained by fabrication limitations. In this study, narrow-bandwidth BG filters based on Ge-Sb-Se chalcogenide materials are investigated. The structure of the filter is designed by optimizing the grating period, corrugation height, and grating number. The large corrugation of chalcogenide BG is more friendly and convenient for manufacturing process. The symmetric and asymmetric corrugation filters are then fabricated and characterized. Experimental results show a half-maximum bandwidth of 0.97 nm and 0.32 nm for symmetric and asymmetric filters, respectively, which demonstrates excellent narrow-bandwidth filtering performance of chalcogenide BG.

A novel coronavirus, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has been identified as the causative agent of the current coronavirus disease 2019 pandemic. Development of animal models that parallel the clinical and pathologic features of disease are highly essential to understanding the pathogenesis of SARS-CoV-2 infection and the development of therapeutics and prophylactics. Several mouse models that express the human angiotensin converting enzyme 2 (hACE2) have been created, including transgenic and knock-in strains, and viral vector-mediated delivery of hACE2. However, the comparative pathology of these mouse models infected with SARS-CoV-2 are unknown. Here, we perform systematic comparisons of the mouse models including K18-hACE2 mice, KI-hACE2 mice, Ad5-hACE2 mice and CAG-hACE2 mice, which revealed differences in the distribution of lesions and the characteristics of pneumonia induced. Based on these observations, the hACE2 mouse models meet different needs of SARS-CoV-2 researches. The similarities or differences among the model-specific pathologies may help in better understanding the pathogenic process of SARS-CoV-2 infection and aiding in the development of effective medications and prophylactic treatments for SARS-CoV-2.

It is of great significance for improving electron transmission performance by changing of the outer ring structure of iron phthalocyanine. Herein, 4 (pyridine-2, 3-yl) iron phthalocyanine (FepyPc), as N-rich pyridyl-iron phthalocyanine derivative, was introduced to degrade pharmaceutical contaminants. The catalytic degradation of organic pollutants with FepyPc was studied by activating peroxymonosulfate (PMS) at room temperature. The results clarified that the removal rate of carbamazepine (CBZ) was close to 100% within 60 min and the calculated apparent rate constant was about 2 times larger than FePc, which proved that FepyPc had superior performance. Four active species were identified for the degradation of CBZ, including superoxide radical (•O2-), singlet oxygen (1O2), sulfate radical (SO4•-) and hydroxyl radical (•OH). In addition, the possible reaction mechanism was inferred in FepyPc/PMS/sunlight system for CBZ removal. Finally, the CBZ degradation pathway was proposed by using ultra-performance liquid chromatography and high definition mass spectrometry (UPLC/HDMS). This research provided a meaningful and efficient method for the elimination of pharmaceutical contaminants.

SARS-CoV-2 is evolving with mutations throughout the genome all the time and a number of major variants emerged, including several variants of concern (VOC), such as Delta and Omicron variants. In this study, we demonstrated that mutations in the regions corresponding to the sequences of the probes and 3'-end of primers have a significant impact on qPCR detection efficiency. We also found that the G28916T mutation of the N gene accounts for 78.78% sequenced genomes of Delta variant. It was found that detection sensitivity of G28916T mutant was 2.35 and 1.74 times less than that of the wt sequence and detection limit was reduced from 1 copy/µl to 10 copies/µl for the commercially available CP3 and CP4 primer/probe sets. These results indicate that the detection probes and primers should be optimized to keep maximal detection efficiency in response to the emergence of new variants.

In this article, the chalcogenide slot waveguide is theoretically studied, and the highest power confinement factors of the slot region and the cladding region are obtained to be 36.3% and 56.7%, respectively. A high-sensitivity chalcogenide slot microring resonator sensor is designed and fabricated by electron-beam lithography and dry etching. The structure increases the sensitivity of the sensor compared with the conventional evanescent field waveguide sensor. The cavity has achieved a quality factor of 1 × 104 by fitting the resonant peaks with the Lorentzian profile, one of the highest quality factors reported for chalcogenide slot microring resonators. The sensor sensitivity is measured to be 471 nm/RIU, which leads to an intrinsic limit of detection of 3.3 × 10--4 RIU.
